背景技術(shù)
促紅細(xì)胞生成素(EPO)是由腎臟合成和分泌的一種生長(zhǎng)因子,能促使紅細(xì)胞生 產(chǎn)和成熟(紅細(xì)胞生成)。最初認(rèn)為EPO僅僅與紅細(xì)胞的生產(chǎn)和成熟有關(guān),但最近的研 究發(fā)現(xiàn)EPO除了具有促進(jìn)紅細(xì)胞生成的作用之外,還有很強(qiáng)的促進(jìn)神經(jīng)元生存(神經(jīng) 保護(hù))的作用。體外實(shí)驗(yàn)表明,EPO可以減輕或抑制由缺糖缺氧NMDA毒性,自由基 等多種因素引起的神經(jīng)元損傷。大量動(dòng)物實(shí)驗(yàn)表明,EPO注射可以減少缺血梗死面積, 同時(shí)可以改善神經(jīng)功能恢復(fù)。最近的一個(gè)臨床實(shí)驗(yàn)結(jié)果令人鼓舞在中風(fēng)5小時(shí)給予 大劑量EPO注射(33000U/次),可以明顯減少由大腦中動(dòng)脈阻塞所致的腦梗死體積, 同時(shí)可以明顯提高病人的祌經(jīng)功能和臨床結(jié)果。這一進(jìn)展意義重大,其一,目前中風(fēng) 唯一有效的藥物是t-PA,其治療窗口為中風(fēng)后3小時(shí)內(nèi),而EPO擴(kuò)大了治療窗口,從 而使更多中風(fēng)患者得到治療;其二, EPO作為新型腦保護(hù)治療試劑可以用于中風(fēng)后神 經(jīng)功能康復(fù)治療。然而,由于腦保護(hù)屏障的作用,只有不到1%EP0可穿過(guò)血腦屏障 而進(jìn)入大腦。因此,需要大劑量注射才內(nèi)能在大腦內(nèi)達(dá)到一定治療濃度,其劑量遠(yuǎn)遠(yuǎn) 高于治療貧血所使用的劑量。同時(shí),EPO的祌經(jīng)保護(hù)作用是通過(guò)激活一系列信號(hào)通道 而實(shí)現(xiàn)的,比如PI3/Akt系統(tǒng),而EPO誘導(dǎo)的PI3/Akt信號(hào)通常僅能維持48小時(shí),因 此需要多次注射來(lái)維持生存信號(hào)的激活。大劑量,多次注射EPO會(huì)產(chǎn)生一連串高風(fēng)險(xiǎn) 的副作用,比如紅細(xì)胞壓積升高而使血液變得粘稠,同時(shí)EPO可刺激血小板的生成。 這些高危因素可導(dǎo)致紅色血栓形成從而增加腦卒中患者發(fā)生再次中風(fēng)的可能。因此, 若能選擇性的降低或去除EPO的促紅細(xì)胞生成作用,而維持其神經(jīng)保護(hù)作用,將帶來(lái) 腦中風(fēng)治療的變革。本發(fā)明即是提供一種新型的無(wú)促紅細(xì)胞生成作用,但有神經(jīng)保護(hù) 作用的新型EPO。
EPO與EPO受體結(jié)合后,激活一系列信號(hào)通路而促進(jìn)細(xì)胞的生成和成熟。在EPO 分子中,有三個(gè)區(qū)域與EPO受體結(jié)合有關(guān),這三個(gè)區(qū)域分別是第11至15號(hào)氨基酸(詳見(jiàn)SEQ ID No.4),第44至51號(hào)氨基酸(SEQ ID No.5)以及第100至108號(hào)氨基酸 (SEQIDNo.6)。研究發(fā)現(xiàn),若在上述三個(gè)區(qū)域中發(fā)生點(diǎn)突變,EPO分子將失去與EPO 受體結(jié)合的能力,從而失去刺激紅細(xì)胞生成的功能。EPO具有神經(jīng)保護(hù)作用,目前的 研究認(rèn)為EPO通過(guò)與神經(jīng)元上的EPO受體結(jié)合,激活與促紅細(xì)胞生成相同的信號(hào)通
路而發(fā)揮神經(jīng)保護(hù)作用。因此,從理論上來(lái)講,破壞EPO受體結(jié)合位點(diǎn)后,EPO將不
再具有祌經(jīng)保護(hù)作用。
因此現(xiàn)階段制備具有神經(jīng)保護(hù)作用無(wú)促紅細(xì)胞作用的EPO是本領(lǐng)域技術(shù)人員研 究的重點(diǎn)。

發(fā)明內(nèi)容
本發(fā)明要解決的技術(shù)問(wèn)題是提供一種具有神經(jīng)保護(hù)作用,但無(wú)促紅細(xì)胞生成特性 的新型促紅細(xì)胞生成素。
本發(fā)明要解決的另一個(gè)技術(shù)問(wèn)題是提供這種具有神經(jīng)保護(hù)作用,但無(wú)促紅細(xì)胞 生成特性的新型促紅細(xì)胞生成素在制藥領(lǐng)域的用途。
為實(shí)現(xiàn)上述目的,本發(fā)明采用以下技術(shù)方案
一種無(wú)促紅細(xì)胞生成特性、具有神經(jīng)保護(hù)作用的促紅細(xì)胞生成素,為野生型人促 紅細(xì)胞生成素在下述三個(gè)區(qū)域中至少一個(gè)區(qū)域進(jìn)行突變而產(chǎn)生的變構(gòu)體
(1) VLQRY:人促紅細(xì)胞生成素的第11-15號(hào)氨基酸;
(2) TKVNFYAW:人促紅細(xì)胞生成素的第44-51號(hào)氨基酸;
(3) SGLRSLTTL:人促紅細(xì)胞生成素的第100-108號(hào)氨基酸。
所述突變?yōu)閱吸c(diǎn)或多點(diǎn)突變、單個(gè)或多個(gè)氨基酸缺失、以及突變加缺失復(fù)合突變。

人類EP0 cDNA(序列W)編碼193個(gè)氨基酸(序列tt2) , EPO初級(jí)蛋白合成后, 其氨基端含27個(gè)氨基酸殘基的信號(hào)肽經(jīng)切割后變?yōu)槌墒霦P0(序列tt3)。因此成熟的 人類EPO為166個(gè)氨基酸。下面所列出的突變型人類促紅細(xì)胞生成素的氨基酸序列均 為成熟后的氨基酸序列。
人類EPO cDNA用PCR方法進(jìn)行定點(diǎn)突變,獲得了 62個(gè)新突變型EPO cDNA。 這些突變主要集中在以下三個(gè)與EPO受體結(jié)合有關(guān)區(qū)域內(nèi)VLQRY(人促紅細(xì)胞生成 素的第11-15號(hào)氨基酸,序列SEQ ID No.4),TKVNFYAW (人促紅細(xì)胞生成素的第44-51 號(hào)氨基酸,序列SEQ ID No.5)和SGLRSLTTL (人促紅細(xì)胞生成素的第100-108號(hào)氨 基酸,序列SEQIDNo.6)。將這些突變后的EPOcDNA裝入真核表達(dá)載體并在人胚腎 293細(xì)胞系中進(jìn)行表達(dá)。通過(guò)篩選穩(wěn)定高表達(dá)細(xì)胞株,我們成功制備了 62個(gè)含有單個(gè)突 變氨基酸的新型突變型EPO(mEPO)融合蛋白,這些mEPO融合蛋白的具體突變位點(diǎn) 如下(其中第一和第三個(gè)字母分別為正常和突變后的氨基酸,兩字母之間的數(shù)字為成 熟促紅細(xì)胞生成素的氨基酸序號(hào),具體氨基酸序列詳見(jiàn)序列表)

我們用體內(nèi)和體外實(shí)驗(yàn)對(duì)這些mEP0是否具有促進(jìn)紅細(xì)胞生成的作用進(jìn)行了研 究,發(fā)現(xiàn)mEP0喪失了維持紅細(xì)胞生成素依賴性細(xì)胞株(32D-EP0R)生存的特性;體內(nèi)實(shí) 驗(yàn)進(jìn)一步證明大劑量、多次注射mEPO不增加小鼠的紅細(xì)胞數(shù)量和血紅蛋白值。這些實(shí) 驗(yàn)數(shù)據(jù)表明mEP0喪失了促進(jìn)紅細(xì)胞生成的能力。令人興奮的是這些突變體和野生型 EP0 —樣,無(wú)論在體內(nèi)還是體外都具有很強(qiáng)的神經(jīng)保護(hù)作用,不僅能夠抑制缺氧和缺 葡萄糖或麗DA毒性有道德祌經(jīng)細(xì)胞死亡的作用,而且體內(nèi)研究發(fā)現(xiàn)mEP0能夠顯著減 少小鼠大腦中動(dòng)脈阻塞模型腦梗死體積。此研究結(jié)果令人意外因?yàn)槟壳暗难芯空J(rèn)為 EPO具有神經(jīng)保護(hù)作用,而神經(jīng)保護(hù)作用是通過(guò)EPO與神經(jīng)元上的EPO受體結(jié)合, 激活與促紅細(xì)胞生成相同的信號(hào)通路而發(fā)揮作用。因此,從理論上來(lái)講,破壞EPO受 體結(jié)合位點(diǎn)后,EPO將不再具有神經(jīng)保護(hù)作用。令人意想不到的是,點(diǎn)突變破壞了 EPO 受體結(jié)合位點(diǎn)后,EPO仍具有神經(jīng)保護(hù)作用,使這種突變型促紅細(xì)胞生成素成為一種 無(wú)促紅細(xì)胞生成作用,但有神經(jīng)保護(hù)作用的新型神經(jīng)保護(hù)藥物。
到目前為止所有治療中風(fēng)的試劑,若在中風(fēng)48小時(shí)后給藥,均無(wú)治療效果。令人 興奮地是新型腦保護(hù)藥物在48小時(shí)后給藥(遲發(fā)性),仍具有明顯的改善神經(jīng)功能的 作用。此結(jié)果意義重大,目前中風(fēng)唯一有效的藥物是t-PA,其治療窗口非常短,為中 風(fēng)后3小時(shí)內(nèi),因此只有不到10%的中風(fēng)患者適合t-PA治療。而EPO擴(kuò)大了治療窗口 , 從而使更多中風(fēng)患者得到治療。此外,新型腦保護(hù)藥物可以長(zhǎng)期注射而對(duì)血液系統(tǒng)沒(méi) 有刺激作用,特別適用于用于腦中風(fēng)的康復(fù)治療。
本發(fā)明的優(yōu)點(diǎn)與效益(1)新型腦保護(hù)藥物具有神經(jīng)保護(hù)作用,但無(wú)促紅細(xì)胞生 成作用,從而避免了常規(guī)促紅細(xì)胞生成素有可能導(dǎo)致的繼發(fā)性血栓形成,增加了臨床
6應(yīng)用的安全性;(2)新型腦保護(hù)藥物擴(kuò)大了治療群體,使血液粘稠度較高而不能用常規(guī) 促紅細(xì)胞生成素進(jìn)行治療的腦卒中患者得到治療;(3)到目前為止,所有治療中風(fēng)的試 劑,若在中風(fēng)48小時(shí)后給藥,均無(wú)治療效果,而新型腦保護(hù)藥物48小時(shí)后給藥,仍 具有明顯的改善神經(jīng)功能的作用。這將使絕大多數(shù)腦卒中患者得到治療;(4)新型促紅 細(xì)胞生成素可以長(zhǎng)期注射而對(duì)血液系統(tǒng)沒(méi)有刺激作用,特別適用于用于腦卒中的康復(fù) 治療。因此這種新型促紅細(xì)胞生成素在腦卒中的治療方面具有良好的臨床應(yīng)用前景-突變型促紅細(xì)胞生成素?cái)U(kuò)大了腦卒中患者治療群體,使更多腦卒中患者得到治療;突 變型促紅細(xì)胞生成素可以長(zhǎng)期注射而對(duì)血液系統(tǒng)沒(méi)有刺激作用,特別適用于腦卒中的 康復(fù)治療。
下面結(jié)合具體實(shí)施方式
對(duì)本發(fā)明作進(jìn)一步說(shuō)明,并非對(duì)本發(fā)明的限定,依照本領(lǐng) 域公知的現(xiàn)有技術(shù),本發(fā)明的實(shí)施方式并不限于此,因此凡依照本公開(kāi)內(nèi)容所作出的 本領(lǐng)域的等同替換,均屬于本發(fā)明的保護(hù)范圍。


圖1-1為mEP0全基因序列示意圖。
圖1-2為部分突變型EP0的DNA測(cè)序結(jié)果。
圖2為表達(dá)質(zhì)粒pcDNA-mEP0-his6的物理圖。
圖3為表達(dá)突變型EPO融合蛋白穩(wěn)定細(xì)胞株的建立及其mEPO融合蛋白的制備 及純化pcDNA-EP0-His6轉(zhuǎn)染293細(xì)胞,用G418篩選高表達(dá)細(xì)胞株。取2ul培養(yǎng)基 上清液用Western blot分析rnEP0的表達(dá)。我們成功地分離純化了 62個(gè)含有單個(gè)突變 氨基酸的新型突變型EPO融合蛋白。圖3所示為S1041-EP0 (序列tt45),其分子量約 34-40 kD(泳道2),與野生型EP0的分子量相似(泳道1)。
圖4為體內(nèi)和體外實(shí)驗(yàn),證明niEP0無(wú)促紅細(xì)胞生成特性(A)應(yīng)用32D-EpoR 細(xì)胞系進(jìn)行體外紅細(xì)胞生成活力檢測(cè)。細(xì)胞(2xl05)在含有1 Li/ml野生型EP0或含 有l(wèi)和100 U/ml mEPO的培養(yǎng)基中進(jìn)行孵育培養(yǎng),72h后進(jìn)行細(xì)胞計(jì)數(shù)。數(shù)據(jù)用平均 值土標(biāo)準(zhǔn)誤表示(n40)。圖中所示為S1041-EP0 (序列tt45),其它序列結(jié)果與此相似。 (B)小鼠活體內(nèi)紅細(xì)胞生成活力檢測(cè)。mEPO或野生型EPO以5000 U/kg總量進(jìn)行 腹腔注射,每周2次,連續(xù)4周注射。每2周進(jìn)行一次血中血紅蛋白濃度檢測(cè)。數(shù)據(jù) 以平均值土標(biāo)準(zhǔn)誤表示,n=8。與野生型EPO注射組相比較*p<0.05, **p<0.01。圖中 突變體S104I-EP0(序列#45),其它序列結(jié)果與此相似。圖5為體外實(shí)驗(yàn),提示mEPO具有抗OGD和NMDA的腦保護(hù)作用(A)為暴露于 薩DA24小時(shí)后顯微鏡下(a-d)和煙酸己可堿染色后形態(tài)學(xué)表現(xiàn)(e-h)。對(duì)照組神經(jīng) 元見(jiàn)圖a和e。 NMDA導(dǎo)致凋亡樣(f圖內(nèi)紅箭頭)和壞死樣細(xì)胞死亡(f圖內(nèi)黃色 箭頭)。EP0(c, g)或mEP0(d, h)預(yù)處理24小時(shí)可減少神經(jīng)元死亡。(B)為,DA 神經(jīng)毒性導(dǎo)致的細(xì)胞死亡計(jì)數(shù)。EP0或mEP0預(yù)處理后暴露于NMDA可明顯減少高 密度細(xì)胞核形態(tài)細(xì)胞數(shù)量。(C)為單獨(dú)醒DA處理后LDH釋放量作為對(duì)照EP0和niEP0 處理的神經(jīng)元釋放LDH的相對(duì)含量。(**〈0. 01 ,數(shù)據(jù)用均值+SEM來(lái)表示,n=12)。

圖7為新生鼠缺氧缺血模型,提示mEP0具有促進(jìn)神經(jīng)功能恢復(fù)的功能出生 后7天的Sprague-Dawley新生鼠給予缺氧缺血130分鐘,然后給予腹腔注射mEPO (5000U/kg),用"腳踩空實(shí)驗(yàn)"檢測(cè)祌經(jīng)功能。圖7A為mEP0注射,祌經(jīng)功能檢測(cè) 及測(cè)定腦梗死體積時(shí)間表;盡管遲發(fā)性mEPO注射不能減少缺氧缺血所致的腦組織(圖 7B和7C),但能顯著提高神經(jīng)功能(圖7D,7E和7F)。
具體實(shí)施例方式
實(shí)施例1: EP0 cDNA定點(diǎn)突變
從人類腎臟cDNA文庫(kù)中擴(kuò)增人類EP0cDNA,用PCR方法進(jìn)行定點(diǎn)突變,獲得了 62個(gè)新突變型EP0 cDNA,并將這些突變后的EPO cDNA裝入真核表達(dá)載體,詳細(xì)步驟為 一、人類EPO cDNA的擴(kuò)增
人類EPOcDNA編碼193個(gè)氨基酸,EPO初級(jí)蛋白合成后,其氨基端含27個(gè)氨 基酸殘基的信號(hào)肽經(jīng)切割后變?yōu)槌墒霦PO。因此成熟的人類EPO為166個(gè)氨基酸。 基于上述研究結(jié)果,將用于分離純化的組氨酸標(biāo)簽(His-6)加于EP0cDNA的3'端。 含有His-6標(biāo)簽的人類EPO cDNA用PCR方法從人類腎臟Marathon-ready cDNA文 庫(kù)(Clontech , Mountain View,美國(guó))中擴(kuò)增。引物設(shè)計(jì)依照己發(fā)表的人類EPO cDNA序列(基因序列號(hào)畫(huà)000799)。其中上游序列含有BamHI及K0ZAK序列(劃 線部分)5' -ACGGATCCGCCACCATGGGGGT GCACGAATGTCTTGC-3,。下游引物序列為5' -/Jg水AATTClr①口rG馬蕩67Y ACCgtcccctgccctgcaggcctcccctg-3', 從3'至5,次含有EPO下游序列(小寫(xiě)部分),六個(gè)組氨酸組成的組氨酸標(biāo)簽(斜 體部分)以及EcoRI克隆位點(diǎn)(方框部分)。在組氨酸標(biāo)簽及EPO之間加入甘氨酸殘 基(黑體部分),便于肽段的自由旋轉(zhuǎn)。PCR反應(yīng)條件為上述兩引物各lOpmol, Marathon cDNA 2m!,Accuprime1M Taq DNA polymerase (Invitrogen, Carlsbad, 美國(guó))1W。 94'C預(yù)變性2分鐘后,94'C變性30秒,62'C退火30秒,68'C延伸50 秒,28個(gè)循環(huán)后用瓊脂糖凝膠電泳回收PCR產(chǎn)物??寺∪階T克隆載體pGEM-T easy (Promega, Madison,美國(guó))。質(zhì)粒擴(kuò)增及酶切鑒定后進(jìn)一步進(jìn)行DNA測(cè)序。mEPO 全基因序列示意圖見(jiàn)圖l-l。 二、 EPO cDNA定點(diǎn)突變

cDNA全基因用BamHI及EcoRI雙酶切后,亞克隆至 pcDNA3. 1(+)表達(dá)載體中(clontech,美國(guó)),命名為pcDNAiEP0-his6。13實(shí)施例2: mEPO融合蛋白制備及純化
用轉(zhuǎn)染試劑Lipofectamine 2000 (Invitrogen,美國(guó))將載體pcDNA-mEP0-his6 導(dǎo)入293細(xì)胞系。轉(zhuǎn)染24h后,1: 12傳代,加入G418 lmg/ml (Invitrogen,美國(guó)) 培養(yǎng)2周后挑選單個(gè)克隆轉(zhuǎn)入6孔板繼續(xù)培養(yǎng)。取10W培養(yǎng)基用Western blot或 稀釋1000倍后用ELISA鑒定mEP0高表達(dá)細(xì)胞株。上清用Western blot和ELISA鑒定 EP0的表達(dá),結(jié)果見(jiàn)圖2。 mEP0高表達(dá)細(xì)胞株大量擴(kuò)增后,收集培養(yǎng)基用Ni-NTA瓊脂 糖層析柱(QIAGEN, Valencia,美國(guó))回收純化mEP0融合蛋白。簡(jiǎn)言之,培養(yǎng)基用NaOH 將其pH值調(diào)至8. 0,加入5M NaCl至終濃度為300mM。
將5ml Ni-NTA瓊脂糖加入層 析柱中,用PBS洗柱兩遍后,將Ni-NTA瓊脂糖加入培養(yǎng)基中。4'C振搖結(jié)合2小時(shí)后, 將Ni-NTA瓊脂糖培養(yǎng)基加入層析柱中,用磷酸鹽緩沖液(50mM NaH2P04, 300mM NaCl, 20mM咪唑,pH8.0)洗柱6遍,然后加入洗脫液(50mM NaH2P04, 300mM NaCl, 250mM 咪唑,PH8.0) 10ml洗脫mEP0融合蛋白。將洗脫的mEPO融合蛋白加入Slide-A-Lyzer 透析盒中(PIERCE, Rockford,美國(guó)),置于1000ml PBS中進(jìn)行透析,每8小時(shí)更換 一次PBS,透析6次后收集蛋白。純化的蛋白經(jīng)0.2陶濾器過(guò)濾后分裝,凍存于-80。C 或冷凍真空抽干備用。SDS-PAGE電泳后,用考馬斯亮蘭染色鑒定蛋白純度。用ELISA 試劑盒(R&D System, Minneapolis,美國(guó))進(jìn)行定量分析。
理論上講未糖基化的mEPO分子量為23kD。 Western blot法確定純化的mEPO融合 蛋白分子量為36至40kD (圖3),表明我們生產(chǎn)的mEPO為糖基化的。用此法提純的 mEP0純度可達(dá)90。/。,濃度高達(dá)50 U/W。
實(shí)施例3:檢測(cè)mEPO是否具有促紅細(xì)胞生成特性
我們用體內(nèi)和體外實(shí)驗(yàn)對(duì)這些niEPO是否具有促進(jìn)紅細(xì)胞生成的特性進(jìn)行了研究。
一、mEPO促紅細(xì)胞生成生物特性的體外分析 32D-EPOR細(xì)胞為小鼠骨髓來(lái)源的淋巴母細(xì)系32D細(xì)胞穩(wěn)定轉(zhuǎn)染EPO受體后形成 的細(xì)胞系,其生存和增殖完全依賴于EPO的存在,被廣泛用于EPO生物學(xué)活特性的體 外分析。用IDEM培養(yǎng)基(Invitrogen)將32D-EPOR細(xì)胞接種于六孔板中,每孔約 2xl()5個(gè)細(xì)胞。饑餓(無(wú)EPO存在)6小時(shí)后分別加入野生型ETO (1U/ml)或mEPO (1 和100 U/ml)或PBS (對(duì)照),72小時(shí)后用胎盤藍(lán)染色,計(jì)數(shù)活細(xì)胞的數(shù)量。在野 生型EPO存在的條件下,32D-EP0R細(xì)胞在72小時(shí)內(nèi)增殖了大約13倍(2.7x107), 而 mEPO和PBS對(duì)照一樣,只有O. 1%細(xì)胞存活,即使增加mEPO濃度100倍后對(duì)32D-EP0R細(xì)胞的存活也沒(méi)有任何影響,表明mEPO喪失了促紅細(xì)胞生成的能力。 二 、 mEP0對(duì)小鼠血紅蛋白濃度的影響
取體重25-30g, 2-3月齡C3H/HEN雌性小鼠(Taconic Laboratories, Taconic, 美國(guó)),以每公斤體重5000單位的劑量腹腔內(nèi)注射mEPO或野生型EP0,每周2次, 連續(xù)注射4周。每2周隱靜脈采血一次,用血紅蛋白儀(HemoCue AB, Angelholm,瑞 典)檢測(cè)血液中血紅蛋白濃度。隱靜脈采血具體操作如下采用氣體麻醉,1.5%異氟 垸混合70%N20和30%02經(jīng)氣管插管行吸入麻醉。將覆蓋在小鼠后肢外側(cè)隱靜脈之上 的毛發(fā)剃除,用右手拇指和食指擠壓覆蓋在外側(cè)隱靜脈之上的皮膚,使隱靜脈充盈, 酒精常規(guī)消毒后采用帶有毛細(xì)軟管的25號(hào)針頭采血。當(dāng)取夠100 " 1靜脈血后迅速拔 出針頭,用棉球按壓穿刺點(diǎn)l-3分鐘促進(jìn)創(chuàng)口止血。C3H/HEN雌性小鼠血紅蛋白的 正常值為14.5土2.0 g/100ml,長(zhǎng)期大劑量mEPO注射對(duì)血紅蛋白沒(méi)有影響 (14. 3±1.8 g/100ml),而野生型EPO顯著增加血紅蛋白的含量,在第二周和第四周 分別達(dá)到注射前的115%和130%,表明mEPO喪失了促紅細(xì)胞生成的能力。
三、結(jié)果
體外實(shí)驗(yàn)發(fā)現(xiàn)mEP0喪失了維持促紅細(xì)胞生成素依賴性細(xì)胞株(32D-EPOR)生存的 特性;體內(nèi)實(shí)驗(yàn)進(jìn)一步證明大劑量,多次注射mEPO不增加小鼠的紅細(xì)胞數(shù)量和血紅 蛋白值,表明mEPO喪失了促紅細(xì)胞生成的能力。本發(fā)明的創(chuàng)造性之一在于新型腦保護(hù) 藥物無(wú)促紅細(xì)胞生成作用,從而避免了常規(guī)促紅細(xì)胞生成素有可能導(dǎo)致的繼發(fā)性血栓 形成,增加了臨床應(yīng)用的安全性。
實(shí)施例4: mEPO腦保護(hù)作用的體外研究
為了確定純化的mEPO蛋白是否具有生物活性,我們驗(yàn)證了其在NMDA毒性誘導(dǎo)的 祌經(jīng)細(xì)胞死亡中的作用。
一、大鼠皮層神經(jīng)元原代培養(yǎng)
按Cao等報(bào)道的方法進(jìn)行(Cao G, J Neurosci. 2001, 21 (13): 4678-90)。簡(jiǎn)言 之,取懷孕16至17天的SD大鼠胚胎,用剪刀將大腦皮層組織剪成約lmni3小塊,用 0.0W胰酶緩沖液37'C消化30分鐘后,加入馬血清滅活胰酶,用吸管吹打后過(guò)濾190g 離心10分鐘后收集細(xì)胞,重懸于Neurobasal培養(yǎng)基(Invitrogen)后種于poly-D-lysine coated 24孔板(3X105)或100mm盤(5X 106)中,同時(shí)加入50XB27 (+)添加 劑(invitrogen),每72小時(shí)更換一次液體。用Neurobasal培養(yǎng)基所得神經(jīng)元側(cè)純度
15為95%。
二、 NMDA毒性模型
生長(zhǎng)至11天的原代培養(yǎng)神經(jīng)元更換新鮮培養(yǎng)基,同時(shí)加入B27 (-)添加劑以及 mEP0或野生型EP0 (lU/ml)繼續(xù)培養(yǎng)。24小時(shí)后加入200幽NMDA,作用15分鐘后更 換為新鮮培養(yǎng)基及B27 (-)添加劑,同時(shí)加入mEP0或野生型EP0。 24小時(shí)后取培養(yǎng)基 上清檢測(cè)LDH活性,作為細(xì)胞死亡的標(biāo)記。同時(shí)用Hoechest染色細(xì)胞核,計(jì)數(shù)濃縮及 片段化的細(xì)胞核比例。
三、 結(jié)果
如圖5所示,mEPO能顯著降低由隨DA誘導(dǎo)的細(xì)胞凋亡及壞死,其效果與商用標(biāo) 準(zhǔn)EP0 (股D System, Minneapolis,美國(guó))相似。表明mEP0和野生型EP0—樣,在 抗函DA所致神經(jīng)毒性方面具有相同的保護(hù)作用。
實(shí)施例5: mEPO對(duì)小鼠大腦中動(dòng)脈阻塞模型腦梗死體積的影響
一、 小鼠短暫局部腦缺血模型的制作
利用單股尼龍縫合線栓塞左側(cè)大腦中動(dòng)脈制作局部腦缺血模型。取體重25-30g , 2-3月齡雄性C57/B6小鼠(The Jackson Laboratory, Bar Harbor,美國(guó)),1. 5%異 氟烷/70°州20/30%02混和氣體氣管插管吸入麻醉。手術(shù)全程動(dòng)物的直腸溫度通過(guò)反饋 恒溫加熱毯維持在37.0土0.5 'C之間,栓塞時(shí)使用鼠尾血壓監(jiān)測(cè)器監(jiān)測(cè)小鼠的平均動(dòng) 脈壓,于栓塞后15分鐘取動(dòng)脈血行血?dú)夥治?。栓?0分鐘后于各指定時(shí)間點(diǎn)拔出線 栓恢復(fù)血供造成再灌注。手術(shù)完畢解除麻醉,小鼠北方回籠內(nèi)飼養(yǎng)在2(TC恒溫的空調(diào) 屋內(nèi)。
二、 梗死體積測(cè)定
梗死體積的測(cè)定采用Yang所描述方法(Stroke. 1994, 25(1) :165-70)。概括如下 于再灌注不同時(shí)間點(diǎn)處死動(dòng)物,快速取出腦組織,冠狀位切片,共6片,每片厚2毫 米,將切好的腦片放入用PBS配置的2y。TTC (氯化2, 3, 5-三苯基四氮唑)溶液中, 37. 5'C水浴20分鐘,然后將其放入4%的多聚甲醛中固定15分鐘后取出拍照。使用圖 像分析系統(tǒng)(MCID, St. Catherine' s,加拿大)。計(jì)算機(jī)圖像分析系統(tǒng)計(jì)算各層腦組 織面積。公式為梗塞面積=非梗塞面積-梗塞側(cè)正常腦組織面積,這樣可以減小腦水 腫對(duì)計(jì)算梗塞面積的影響。總的梗塞體積^各層梗塞面積X層厚(2mm)。為了驗(yàn)證TTC 染色測(cè)定梗死體積的準(zhǔn)確性,實(shí)驗(yàn)室進(jìn)行了組織學(xué)染色與之對(duì)比。將動(dòng)物處死后快速取腦,將腦組織放入2-甲基-丁烷在3(TC下冰凍。行冠狀位連續(xù)冰凍切片,每片厚20陶, 每隔0. 5mm取一張腦片(每個(gè)腦組織大概可取22-24張腦片)。切好的腦組織切片進(jìn)行 甲酚紫染色,按上述的方法進(jìn)行梗塞體積的計(jì)算。 三、結(jié)果
我們進(jìn)一步證實(shí)了 mEPO在小鼠腦缺血再灌注模型中的神經(jīng)保護(hù)作用。C57/B6小 鼠大腦中動(dòng)脈栓塞60分鐘再灌注72小時(shí)后造成同側(cè)半球梗死,TTC染色顯示梗死體 積大約為32誦3。缺血后再灌注即刻使用5000U/kg mEPO可明顯減少梗死體積(P〈0. 01, 與空白對(duì)照組比較),其效果與野生型EPO相似(圖6A和6B),同時(shí)可以明顯改善神 經(jīng)功能(圖6C)。表明無(wú)論是體內(nèi)或體外,mEPO都具有很強(qiáng)很的神經(jīng)保護(hù)作用。此研究 結(jié)果令人意外因?yàn)槟壳暗难芯空J(rèn)為EPO具有神經(jīng)保護(hù)作用,而神經(jīng)保護(hù)作用是通過(guò) EPO與神經(jīng)元上的EPO受體結(jié)合,激活與促紅細(xì)胞生成相同的信號(hào)通路而發(fā)揮作用。 因此,從理論上來(lái)講,破壞EPO受體結(jié)合位點(diǎn)后,EPO將不再具有神經(jīng)保護(hù)作用。令 人意想不到的是,點(diǎn)突變破壞了 EPO受體結(jié)合位點(diǎn)后,EPO仍具有神經(jīng)保護(hù)作用,使 這種突變型促紅細(xì)胞生成素成為一種無(wú)促紅細(xì)胞生成作用,但有神經(jīng)保護(hù)作用的新型 神經(jīng)保護(hù)藥物。
實(shí)施例6: mEP0促進(jìn)神經(jīng)功能恢復(fù)
到目前為止所有治療中風(fēng)的試劑,若在中風(fēng)48小時(shí)后給藥,均無(wú)治療效果。令人 興奮地是新型腦保護(hù)藥物在48小時(shí)后給藥(遲發(fā)性),仍具有明顯的改善神經(jīng)功能的 作用。遲發(fā)性給藥仍具有治療效果,其機(jī)制目前尚不清楚,可能與新型腦保護(hù)藥物誘 導(dǎo)血管再生和神經(jīng)再生,從而促進(jìn)神經(jīng)功能的恢復(fù)有關(guān)。為了證實(shí)新型腦保護(hù)藥物是 否通過(guò)誘導(dǎo)血管再生和神經(jīng)再生而發(fā)揮作用,我們進(jìn)一步研究了新型腦保護(hù)藥物對(duì) 新生鼠缺氧缺血模型所致的神經(jīng)功能損傷影響。 一、新生鼠缺氧缺血模型
應(yīng)用出生后7天的新生Sprague-Dawley大鼠(Charles River實(shí)驗(yàn)室,Wilmington, Mass)制備130分鐘缺氧/再灌注模型。簡(jiǎn)而言之,在3%異氟烷麻醉下,結(jié)扎左側(cè)頸 總動(dòng)脈,90分鐘后將動(dòng)物放置于缺氧環(huán)境中(含氧量為8%)。 40分鐘后將動(dòng)物放放回 正常環(huán)境中侍養(yǎng)。非缺血?jiǎng)游镒鳛閷?duì)照組。將野生型rEP0和變構(gòu)型mEP0溶于0. 1%牛 血清白蛋白磷酸鹽緩沖溶液中,按照?qǐng)D7A所示劑量腹腔注射制備野生型rEP0 (5000 U/kg)和變構(gòu)型mEP0 (5000U/kg)。 0. 1%牛血清白蛋白磷酸鹽緩沖溶液注射作為非治療對(duì)照組。
二、 神經(jīng)功能檢測(cè)
我們應(yīng)用"腳踩空實(shí)驗(yàn)"檢測(cè)神經(jīng)功能。簡(jiǎn)言之,動(dòng)物被放置在升高8.5毫米 的網(wǎng)格地板上(34 x 29 mm)。網(wǎng)眼面積為2. 2 x 2. 2 cm。當(dāng)大鼠在網(wǎng)格上移動(dòng)的時(shí) 候,未受損傷的動(dòng)物可以將爪子放在網(wǎng)架,在網(wǎng)上移動(dòng)時(shí)可以抓牢格子。缺氧損傷的 動(dòng)物不能準(zhǔn)確地將一側(cè)前或者后肢放置踩在格子上,甚至?xí)⑶爸笾珡木W(wǎng)眼中跌落, 即為1次"腳踩空"。觀察1分鐘,記錄腳步的總數(shù),"腳踩空"次數(shù)。圖7D顯示缺 氧/再灌注之后感覺(jué)運(yùn)動(dòng)行為顯著缺失。與對(duì)照組相比,非治療組動(dòng)物對(duì)側(cè)的(圖7E) 和同側(cè)的(圖7F)肢體"腳踩空"次數(shù)均顯著增高,EPO的應(yīng)用可以減小這種神經(jīng)功 能缺失的程度(圖7D, 7E和7F)(圖6B-D)。因此,外源性EP0在改善缺氧/再灌注后 的神經(jīng)損傷是有效的。
三、 結(jié)果
出生后7天的Sprague-Dawley新生鼠給予缺氧缺血130分鐘,2, 4, 6, 9和 13天后分別給予腹腔注射mEPO (5000U/kg),然后檢測(cè)祌經(jīng)功能的恢復(fù)。我們發(fā)現(xiàn)盡 管遲發(fā)性mEPO注射不能減少缺氧缺血所致的梗死體積(圖7B和7C),但能顯著提高 神經(jīng)功能的恢復(fù)(圖7D, 7E和7F),其效果與野生型EP0相似,而且mEPO可以大劑 量多次注射而不用擔(dān)心野生型EPO所致的血液學(xué)副作用。此結(jié)果意義重大,目前中風(fēng) 唯一有效的藥物是t-PA,其治療窗口非常短,為中風(fēng)后3小時(shí)內(nèi),因此只有不到10% 的中風(fēng)患者適合t-PA治療。而EPO擴(kuò)大了治療窗口,從而使更多中風(fēng)患者得到治療。 此外,新型腦保護(hù)藥物可以長(zhǎng)期注射而對(duì)血液系統(tǒng)沒(méi)有刺激作用,特別適用于用于腦 中風(fēng)的康復(fù)治療。序列表

2. 根據(jù)權(quán)利要求1所述的無(wú)促紅細(xì)胞生成特性、具有神經(jīng)保護(hù)作用的促紅細(xì)胞生成素, 其特征在于所述突變?yōu)閱吸c(diǎn)或多點(diǎn)突變、單個(gè)或多個(gè)氨基酸缺失、以及突變加缺失復(fù)合突 變。
3. 根據(jù)權(quán)利要求1或2所述的無(wú)促紅細(xì)胞生成特性、具有神經(jīng)保護(hù)作用的促紅細(xì)胞生 成素,其特征在于所述無(wú)促紅細(xì)胞生成特性的新型促紅細(xì)胞生成素為序列表SEQID No.7至SEQ ID No.68中的任一氨基酸序列。
4. 根據(jù)權(quán)利要求3所述的無(wú)促紅細(xì)胞生成特性、具有神經(jīng)保護(hù)作用的促紅細(xì)胞生成素, 其特征在于,特別是第14號(hào)精氨基突變?yōu)楸被?序列#14),谷氨酰胺(序列S15)以 及谷氨酸(序列#16);第45號(hào)賴氨酸突變?yōu)楸被?序列#21)以及谷氨酸(序列 #22);第100號(hào)絲氨酸突變?yōu)楸被?序列tt33)以及谷氨酸(序列#34);第103號(hào)精 氨基突變?yōu)楸被?序列tt39)以及谷氨酸(序列tt40);第104號(hào)絲氨酸突變?yōu)楸被?序 列M4),異亮氨酸(序列M5)以及谷氨酸(序列1146)。
5. —種無(wú)促紅細(xì)胞生成特性、具有神經(jīng)保護(hù)作用的促紅細(xì)胞生成素,其特征在于 除權(quán)利要求l中所述三個(gè)區(qū)域外,在人促紅細(xì)胞生成素的第6, 7, 10, 20, 21, 29, 33, 38, 42, 52, 59, 63, 67, 70, 83, 96, 97, 126, 142, 149, 150, 151, 152, 153, 155, 156, 161號(hào)氨基酸位置進(jìn)行突變而產(chǎn)生的變構(gòu)體。
6. 根據(jù)權(quán)利要求1或5所述的無(wú)促紅細(xì)胞生成特性的新型促紅細(xì)胞生成素,與其 它蛋白或多肽進(jìn)行融合、置換、化學(xué)交聯(lián)而產(chǎn)生的新型融合蛋白。
7. —種無(wú)促紅細(xì)胞生成特性、具有神經(jīng)保護(hù)作用的促紅細(xì)胞生成素在制備保持、 增強(qiáng)神經(jīng)保護(hù)作用、或用于神經(jīng)功能康復(fù)治療的藥物中的應(yīng)用。
8. —種無(wú)促紅細(xì)胞生成特性、具有神經(jīng)保護(hù)作用的促紅細(xì)胞生成素在制備用于中 樞神經(jīng)系統(tǒng)以外的臟器,如心臟、腎臟等臟器的缺血性損傷的治療藥物中的應(yīng)用。
全文摘要
本發(fā)明涉及一種無(wú)促紅細(xì)胞生成特性、具有神經(jīng)保護(hù)作用的促紅細(xì)胞生成素及其用途。本發(fā)明為在野生型人促紅細(xì)胞生成素的下述三個(gè)區(qū)域中的至少一個(gè)區(qū)域及其它28個(gè)散在位點(diǎn)發(fā)生突變產(chǎn)生的變構(gòu)體(1)VLQRY人促紅細(xì)胞生成素的第11-15號(hào)氨基酸;(2)TKVNFYAW人促紅細(xì)胞生成素的第44-51號(hào)氨基酸;(3)SGLRSLTTL人促紅細(xì)胞生成素的第100-108號(hào)氨基酸。本發(fā)明具有神經(jīng)保護(hù)作用,但無(wú)促紅細(xì)胞生成作用,避免了常規(guī)促紅細(xì)胞生成素導(dǎo)致繼發(fā)性血栓形成,增加臨床應(yīng)用安全性;擴(kuò)大了治療群體,使血液粘稠度較高而不能用常規(guī)促紅細(xì)胞生成素進(jìn)行治療的腦卒中患者得到治療;本發(fā)明48小時(shí)后給藥,仍具有明顯改善神經(jīng)功能作用??梢蚤L(zhǎng)期注射對(duì)血液系統(tǒng)沒(méi)有刺激。
