背景技術(shù):
微孢子蟲是普遍分布于自然界的細(xì)胞內(nèi)專性寄生原蟲,近150個(gè)屬1200余種,有著 極為廣泛的宿主,包括脊椎動物和無脊椎動物,是經(jīng)濟(jì)昆蟲、魚類、兔類、產(chǎn)毛動物、嚙 齒類及靈長類的致命病原。而家蠶微孢子蟲(7fow歷s ^歷Z^cis),分類地位屬于原生動物 門,單絲孢子蟲亞門,微孢子蟲綱。由微孢子蟲寄生于經(jīng)濟(jì)昆蟲家蠶而引起嚴(yán)重的傳染性 微粒子病,且由于微孢子原蟲可以通過胚胎向下代傳播, 一直被養(yǎng)蠶國家和地區(qū)列為蠶業(yè) 生產(chǎn)法定檢疫對象。單就我國每年因帶毒種和微粒子病而帶來的直接經(jīng)濟(jì)損失即達(dá)上千萬 元以上。攻克微粒子病這一蠶絲業(yè)的頑癥,是蠶業(yè)界的重點(diǎn)和難點(diǎn)研究領(lǐng)域。
微粒子病是由微孢子蟲寄生宿主而引起的疾病。微孢子蟲以其獨(dú)特的侵染方式,廣泛 寄生并感染極具經(jīng)濟(jì)價(jià)值的昆蟲、魚類、嚙齒類、兔類、皮毛動物、靈長類以及家庭寵物 如狗和鳥,引發(fā)嚴(yán)重疾病甚至死亡,嚴(yán)重制約著世界蠶業(yè)生產(chǎn)和動物保育。甚至還能機(jī)會 性地感染艾滋病(HIV)患者、器官移植病人、服用免疫抑制劑藥物病人和先天性免疫機(jī) 能不健全者,引起呼吸系統(tǒng)、泌尿系統(tǒng)和皮膚潰爛等惡性疾病,加劇病情,加速病人的死 亡。100多年來,盡管己使用了多種化學(xué)藥物進(jìn)行治療,如咪唑類、煙曲霉素、歌咪啶 堿、苯萊特、甲基托布津等等,但治療效果并不理想,至今仍未發(fā)現(xiàn)有治療微孢子蟲病的 特效藥物。
極管是家蠶微孢子蟲繁殖和侵染的重要器官,在家蠶微孢子蟲對家蠶的侵染中起著關(guān) 鍵性作用。極管長度一般在50 15(Vm,直徑一般在O. 1 0.2pm范圍。極管具有柔韌性, 在運(yùn)送孢原質(zhì)的過程中其管直徑可以增加到0.4pm,孢原質(zhì)運(yùn)送完畢后其長度會縮短5 10%。在家蠶微孢子蟲入侵過程中,極管在適當(dāng)剌激后迅速彈出并"刺入"家蠶細(xì)胞中, 同時(shí)將孢原質(zhì)注入其細(xì)胞質(zhì),完成對家蠶的入侵。而其組成成分主要是蛋白,而極管蛋白 PTP1是它主要的組成部分。極管蛋白PTP1是一種伴刀豆球蛋白,是極管的主要蛋白,在 £力e^o歷中PTP1蛋白的含量占整個(gè)極管蛋白的70%。通過SDA-PAGE和銀染分析PTP1發(fā) 現(xiàn),不同種類的微孢子蟲的PTP1分子大小不等。對不同微孢子蟲PTP1蛋白的研究表明,PTP1蛋白具有疏水性、高脯氨酸含量和免疫抗原性等諸如特征(Delbac et al.,1998)。 而PTP2和PTP3也是構(gòu)成極管的重要組分。有研究認(rèn)為,在極管彈出過程中,PTP1和PTP2 在孢子激活之前呈類結(jié)晶狀蛋白質(zhì)單體,發(fā)芽過程中在極管頂端形成聚合體,而PTP3通 過調(diào)控PTP1-PTP2聚合體的構(gòu)象狀態(tài)影響極管的彈出。另外,極管蛋白PTP1的糖基化作 用對極管的構(gòu)造起著重要的作用,從而間接影響微孢子蟲對宿主細(xì)胞的入侵。但是目前關(guān) 于微孢子蟲極管蛋白的研究僅局限于^7c印/ a"'tozo朋、67收朋、^7es朋和7fe^歷a等部 分微孢子蟲屬的少量微孢子蟲。
發(fā)明內(nèi)容
本發(fā)明擬基于基因組分析數(shù)據(jù),其目的在于提供一種家蠶微孢子蟲極管蛋白PTP3基 因。通過分析PTP3的功能、組成和特性,弄清極管蛋白PTP3在家蠶微孢子蟲極管彈出過 程中的作用,探明家蠶微孢子蟲侵染宿主家蠶致病的機(jī)制,對生產(chǎn)實(shí)踐和疾病防治具有極 其重要的意義。
本發(fā)明申請人通過對家蠶微孢子蟲極絲蛋白PTP3基因的克隆,得到了該蛋白基因的
編碼序列,具體方法如下
(1) 同源性預(yù)測通過對家蠶微孢子蟲與兔腦炎微孢子蟲基因組的基因同源性分析,
得到了家蠶微孢子蟲極管蛋白PTP3基因序列CDS;
(2) 引物設(shè)計(jì)利用引物設(shè)計(jì)軟件Primer 5.0進(jìn)行引物設(shè)計(jì),在正向引物加入BamH
I酶切位點(diǎn),在反向引物加入SalI酶切位點(diǎn),引物序列分別為 PTP3-Primer-F5'-CGGGATCCCTACACATTATTTG-3'
PTP3-Primer-R5-CGGTCGACGTGATGATGGGATTCC-3'
(3) 極絲蛋白PTP3PCR擴(kuò)增和克隆利用提取的微孢子基因組直接作為模板,采 用基因組PCR擴(kuò)增目的片段;將擴(kuò)增片段克隆到pMD18-T (TaKaRa)載體上,然后進(jìn)行
序列測定(上海英俊),從而獲得的極絲蛋白PTP3全長編碼序列;
(4) 序列同源性分析同源性比對在NCBI站點(diǎn)上用BLAST完成,BLAST分析表明家
蠶微孢子蟲極絲蛋白PTP3與NCBI蛋白質(zhì)庫中蛋白質(zhì)沒有顯著的同源性,而極絲蛋白PTP3
是家蠶微孢子蟲極管的重要蛋白,因此極絲蛋白PTP3基因是一種新發(fā)現(xiàn)的蛋白基因。

(1) 由于微孢子蟲極管蛋白PTP3是組成微孢子蟲極管的主要蛋白,而極管是微孢子蟲繁殖和侵染的重要器官,通過它廣泛寄生并感染極具經(jīng)濟(jì)價(jià)值的昆蟲、魚類、嚙齒類、兔類、皮毛動物、靈長類以及家庭寵物如狗和鳥,引發(fā)嚴(yán)重疾病甚至死亡,嚴(yán)重制約著世界蠶業(yè)生產(chǎn)和動物保育。甚至還能機(jī)會性地感染艾滋病(HIV)患者、器官移植病人、服用免疫抑制劑藥物病人和先天性免疫機(jī)能不健全者,引起呼吸系統(tǒng)、泌尿系統(tǒng)和皮膚潰爛等惡性疾病,加劇病情,加速病人的死亡。齒此所獲得的極管蛋白PTP3對研究微孢子蟲繁殖和侵染機(jī)制有其重要作用。通過分析PTP3的功能、組成和特性,弄清極管蛋白PTP3在家蠶微孢子蟲極管彈出過程中的作用,探明家蠶微孢子蟲侵染宿主家蠶致病的機(jī)制,對生產(chǎn)實(shí)踐和疾病防治具有極其重要的意義。
(2) RNAi干涉等試驗(yàn)分析表明,把極絲蛋白PTP3干涉到后,微孢子蟲失去感染宿主的能力。所以微孢子蟲的極絲蛋白PTP3和它潛在的調(diào)控基因?qū)⒊蔀橹委熚㈡咦酉x感染新的因素。還可以通過轉(zhuǎn)基因的手段,使之它高豐度的表達(dá),從而殺害蝗蟲等部分害蟲。
具體實(shí)施方式

應(yīng)用Glimmer3.0軟件,通過對家蠶微孢子蟲與兔腦炎微孢子蟲基因組的基因同源性分析,得到了家蠶微孢子蟲極管蛋白PTP3基因序列CDS;應(yīng)用Primer5. 0軟件,設(shè)計(jì)出一對特異性引物

3、 PTP3基因在制備殺蟲劑特別是殺蝗劑中的應(yīng)用。
全文摘要
一種家蠶微孢子蟲的繁殖和侵染有關(guān)的重要蛋白基因PTP3,依據(jù)家蠶微孢子蟲的9倍基因組數(shù)據(jù),得到編碼家蠶微孢子蟲極管蛋白PTP3基因序列CDS,并經(jīng)一系列生物技術(shù)克隆證實(shí)。本發(fā)明所獲得的極管蛋白PTP3對研究微孢子蟲繁殖和侵染機(jī)制有其重要作用。從而對生產(chǎn)實(shí)踐和微孢子蟲病的防治發(fā)揮作用。RNAi干涉等試驗(yàn)分析表明,把極絲蛋白PTP3干涉到后,微孢子蟲失去感染宿主的能力。所以微孢子蟲的極絲蛋白PTP3和它潛在的調(diào)控基因?qū)⒊蔀橹委熚㈡咦酉x感染新的因素。還可以通過轉(zhuǎn)基因的手段,使之它高豐度的表達(dá),從而殺害蝗蟲等部分害蟲。
